上海药物研究所李亚平研究员、尹琦研究员团队在Advanced Materials(IF=27.4)上发表题为“Personalized Multi-Epitope Nanovaccine Unlocks B Cell-Mediated Multiple Pathways of Antitumor Immunity(个性化纳米疫苗激活B细胞介导抗肿瘤免疫应答)”的研究。研究团队构建了一种基于肿瘤细胞膜囊泡的个性化纳米疫苗,称为CM-CpG-aCD40,通过向B细胞提供多重刺激信号,全面触发抗肿瘤固有的细胞和体液免疫应答,从而实现高效的抗肿瘤作用。
在肿瘤免疫治疗盛行的时代,B淋巴细胞逐渐从“旁观者”转变为“参与者”。尽管增强先天和适应性细胞免疫反应主要是肿瘤免疫治疗的重点,并取得了一定的临床效益,但现有的免疫疗法存在一定缺陷,这使得开发新的免疫激活途径变得十分迫切。B细胞参与体液免疫和细胞免疫,是许多实体肿瘤中第二丰富的淋巴细胞,也是三级淋巴结构(TLS)的重要组成部分。近期的研究表明,B细胞既可以分泌肿瘤特异性抗体,也可以向T细胞提呈抗原,同时参与抗肿瘤细胞免疫和体液免疫。虽然对B细胞表位疫苗在癌症治疗中的研究落后于T细胞疫苗,但B细胞受体(BCR)更敏感,抗原识别能力更强,为开发以B细胞为靶的肿瘤疫苗提供了理论基础。
基于现有研究背景,研究团队构建了一种自体TNBC细胞衍生的疫苗,用于双重激活B细胞和T细胞,并命名为CM-CpG-aCD40。CM-CpG-aCD40是一种个性化多表位肿瘤疫苗,通过在肿瘤细胞膜囊泡上使用小分子交联剂偶联两种免疫增强剂——激动性CD40抗体(aCD40)和CpG制备而成。CM-CpG-aCD40能在淋巴结和脾脏中稳定积累并激活B细胞及树突状细胞。CM-CpG-aCD40通过模拟T细胞依赖和独立的途径激活B细胞,促进抗体产生和细胞免疫。此外,它还能促使巨噬细胞转变为M1型,增强先天免疫反应。联合使用CM-CpG-aCD40和抗PD-L1抗体能显著提高治疗效果,抑制肿瘤生长。该纳米疫苗为基于B细胞的免疫治疗提供了一个有效的平台。
在该项研究中,使用了欣博盛生物(NeoBioscience Technology Co, Ltd)的Rat IgG ELISA 试剂盒、Mouse IgG, IgG1, IgG2a, IgG2b, IgG3和IgM ELISA试剂盒,用于检测脾细胞培养上清液中相关指标的含量。
