近期,华东理工大学刘昌胜院士和王靖教授团队在Bioactive Materials(IF=18.0)上发表题为“Immunoregulative coating for scarless healing in anterior cruciate ligament reconstruction(免疫调节涂层在前交叉韧带重建无瘢痕愈合中的应用)”的文章,在该项研究中,研究团队开发了一种多层免疫调节水凝胶涂层,用于无瘢痕的PET(Polyethylene terephthalate)-骨整合。这种多层炎症调节水凝胶涂层的应用不仅可以去除瘢痕组织屏障,还可以通过增强血管生成和成骨来改善移植物-骨骼整合。此外,它避免了外源性生长因子的过度使用和潜在的并发症,提供了一种更方便可行的治疗策略。

SCS@PGH/PET水凝胶涂层在研究中显示了卓越的特性,包括为细胞渗透而优化的多孔结构,以及增加的对细胞粘附和增殖至关重要的润滑性。再结合硫酸化多糖和生物活性分子已被证明在调节巨噬细胞(Mφs )从促炎性M1状态到再生性M2状态的极化中至关重要(调节了如TGF-β这样的细胞因子),而这种转变对于减少瘢痕形成、促进愈合环境和加速血管生成至关重要。通过体外和体内研究,团队确认了水凝胶对组织修复和再生的显著影响。总体而言,SCS@PGH/PET水凝胶涂层的发现为再生医学中合成韧带的设计和改进提供了坚实的科学基础。
