心肌梗死(MI)是世界范围内导致死亡和残疾的主要原因之一,炎症反应在MI修复中起着关键作用。心肌梗死后,大量中性粒细胞、单核细胞和巨噬细胞在受损心肌募集,以诱导炎症反应。这些炎症细胞的持续浸润阻碍组织修复,进而导致不良重塑和心功能受损。内源性中性粒细胞凋亡的精确调节和诱导可能有利于促进再生。然而,中性粒细胞固有的存活/凋亡平衡是由复杂的信号因子网络监测的,诱导中性粒细胞凋亡的治疗可能会干扰固有免疫反应。而且,在原位精确地实现中性粒细胞凋亡的诱导是很困难的。因此,以凋亡中性粒细胞来克服复杂炎症微环境的替代疗法也许可以有效治疗心肌梗死。


近期,第四军医大学金岩、刘世宇教授和西安交通大学陈鑫教授团队在期刊《Bioactive Materials》上发表了题为“Engineered neutrophil apoptotic bodies ameliorate myocardial infarction by promoting macrophage efferocytosis and inflammation resolution”的文章,构建了工程化中性粒细胞凋亡小体(eNABs)来模拟自然中性粒细胞凋亡,调节炎症反应并增强MI修复。eNABs是通过结合具有良好炎症倾向性和免疫调节特性的自然中性粒细胞凋亡体膜和负载有5-氨基乙酰丙酸己酯盐酸盐(HAL)的介孔二氧化硅纳米颗粒制备的。eNABs主动靶向巨噬细胞和组装的HAL的同时启动血红素生物合成途径,在细胞内释放后产生抗炎胆红素,从而进一步增强抗炎作用。在体内研究中,eNABs有效调节梗死区域的炎症反应以改善心功能。这项研究证明了一种有效的仿生构建策略来调节巨噬细胞的功能以进行MI修复。


本文中使用了欣博盛大鼠TNF-α, IL-6, TGF-β and IL-10 ELISA试剂盒,检测了大鼠巨噬细胞培养上清中细胞因子的含量。
