缺血性心脏病是由于冠状动脉疾病引起的心肌变性、坏死和纤维化,并导致严重左室功能障碍的一种疾病,是导致心力衰竭常见的原因,通过成功开通闭塞的冠状动脉治疗心肌梗死的可显著降低死亡率,因此,学者针对左心室病理性重塑进行了大量研究。心室重塑与心脏功能和结构的变化有关,临床和实验研究都表明炎症反应参与心肌梗死后心室重塑,清除梗死区域死亡细胞和细胞外基质碎片,然而,过度活跃的炎症信号会加剧心肌损伤并导致不良的心室重塑。近日,华中科技大学同济医学院附属协和医院心血管内科夏霓和程翔教授团队在期刊《Basic research in cardiology》(IF: 17.165)上发表了题为“Regulatory B cells improve ventricular remodeling after myocardial infarction by modulating monocyte migration”的文章,旨在确定调节性B细胞(Bregs)对心室重构的影响,并探讨其相关机制。本研究发现过继转移Bregs可改善小鼠心肌梗死模型中的心室重构,如改善心脏功能、减少瘢痕大小和减轻间质纤维化,但不改变存活率。从机制上讲,Bregs降低了单核细胞中C-C基序趋化因子受体2(CCR2)的表达,这抑制了促炎性单核细胞从外周血向心脏募集和骨髓动员。用白细胞介素10(IL-10)抗体治疗可消除Breg介导的心肌梗死保护作用。最后,IL-10中和作用改变了Bregs对单核细胞迁移和CCR2表达的影响。目前的研究表明,通过减少CCR2介导的单核细胞募集和动员,Bregs在限制心肌梗死后心室重构方面具有治疗价值。
本文中使用了欣博盛小鼠IL-10和TGF-β1 ELISA试剂盒,检测了小鼠脾脏组织中IL-10的表达量以及小鼠脾脏调节性B细胞和对照B细胞中IL-10和TGF-β1的表达情况。
