高水平的血管生成与许多类型的癌症的不良预后有关,然而目前存在的抗血管生成疗法的疗效仍不令人满意,尤其是在延长TNBC(triple-negative breast cancer)患者的总生存期方面没有显著效果。研究发现肿瘤相关内皮细胞通过诱导凋亡限制抗肿瘤免疫,通过Fas配体(FasL)介导的机制浸润T淋巴细胞。最近,中国科学院上海药物研究所研究团队在期刊《Advanced Materials》(IF=29.4)上发表题为“Inhibiting Endothelial Cell‐Mediated T Lymphocyte Apoptosis with Integrin‐Targeting Peptide‐Drug Conjugate Filaments for Chemoimmunotherapy of Triple‐Negative Breast Cancer.”的文章,该研究团队设计了一种PDCs(peptide-drug conjugate),通过将7-乙基-10-羟基喜树碱(SN38)通过一个RGDR或HKD基序的亲水片段连接到在其C-末端谷胱甘肽反应接头,PDCs在水溶液中自发组装成细丝解决方案。含1% SN38-RGDR(SN38-HKD/RGDR)的PDC长丝有效靶向表达上调的三阴性乳腺癌(TNBC)细胞和TECs,并诱导肿瘤细胞的免疫原性细胞死亡(ICD)和FasL下调 TECs。SN38-HKD/RGDR增加CD8+ T细胞的浸润、活性和生存力, 从而抑制原发性肿瘤和肺转移瘤的生长。这项研究指出癌细胞和TECs与整合素靶向的协同调节 PDC细丝作为TNBC化学免疫疗法的一种有前途的策略。


本文中使用了欣博盛Mouse VEGF, IL-10, IFN-γ, TNF-α 和 IL-12p70 ELISA 检测了细胞培养液上清中目标蛋白的含量。
