吉西他滨加顺铂(GP)化疗是鼻咽癌(NPC)的一种标准治疗方式。然而,治疗的机制尚不清楚。最近,中山大学科研团队在期刊《Nature Medicine》(IF=82.9)上发表题为“The tumor immune microenvironment of nasopharyngeal carcinoma after gemcitabine plus cisplatin treatment.”的文章,使用单细胞RNA测序,对首次治疗和GP治疗后的NPC样本(n = 15对)的T细胞和B细胞受体测序匹配,结果显示GP化疗会激活先天样B细胞(ILB)主导的抗肿瘤免疫反应。化疗诱发的DNA片段激活了STING, 增加干扰素依赖途径在癌细胞中表达MHC Class I,并同时诱导 ILB通过Toll样受体9信号通路。对 germinal centers无效的化疗,ILB通过ICOSL–ICOS轴进一步扩大了follicular helper和helper type 1 T 细胞随后增强三级淋巴器官样结构中的细胞毒性T细胞。ILB频率是与3期临床试验中的接受GP化疗的NPC患者(NCT01872962,n = 139)总体无病生存率呈正相关。ILB频率可以作为GP和免疫疗法联合治疗(n = 380)以下治疗的NPC患者中作为有利结果的预测因子。该研究提供了GP化疗后肿瘤免疫微环境的高分辨率地图并发现B细胞为中心的抗肿瘤作用。研究还确定并验证了在NPC以全科医生为基础的治疗中ILB作为一个潜在的生物标志物可以改善病人管理。


本文中使用了欣博盛Human IFN-β ELISA kit检测了细胞培养上清中该蛋白的含量。
