合成生物学的快速发展激发了人们对能够诊断和治疗疾病的工程微生物的兴趣。然而,设计检测动态疾病相关生物标志物的细菌,然后进行驱动治疗仍然很困难。最近,华东理工大学的研究团队在期刊《Cell Host & Microbe》(IF=31.316)上发表题为“Biomarker-responsive engineered probiotic diagnoses, records, and ameliorates inflammatory bowel disease in mice.”的文章,他们开发了一种工程益生菌,它可以非侵入性地实时监测和记录炎症性肠病(inflammatory bowel disease,IBD)的发生和发展,并可以通过一种自我调节的机制来响应这些生物标志物,从而释放治疗药物。这些用于诊断和治疗的智能响应细菌(i-ROBOT)由大肠杆菌Nissle 1917组成,该细菌通过激活碱基编辑系统来产生可遗传的基因组DNA序列并产生比色信号,从而对炎症标记物硫代硫酸盐的水平做出响应。硫代硫酸盐的波动也驱动免疫调节剂AvCystatin的可调释放。给患有结肠炎的小鼠口服i-ROBOT,在处理过的粪便和结肠样品中产生分子记录信号,并有效改善疾病。i-ROBOT为胃肠和其他代谢疾病提供了一个有前景的设计思路。


本文中使用了欣博盛Human IL-10和IL-12/23 p40 ELISA kits检测了细胞培养液上清样本中这些细胞因子的含量。
