近期，中国科学院上海药物研究所于海军老师课题组在国际权威学术期刊Advanced materials上发表了题为“Engineering Stimuli-Activatable Boolean Logic Prodrug Nanoparticles for Combination Cancer Immunotherapy”的论文，本文首次报道了一种用于免疫调节剂（如免疫激活剂和免疫抑制剂）的肿瘤靶向协同输送及联合免疫治疗的布尔逻辑前药纳米粒（BLPNs）。构建了一个刺激激活BLPNs库，通过调整输入组合包括肿瘤微环境中的细胞外基质金属蛋白2/9（MMP-2/9）、细胞内酸度（pH=5.0-6.0）和还原型谷胱甘肽，来产生YES/AND逻辑输出。通过指定正交刺激-不稳定部分的分子结构和连接性，同时利用肿瘤部位内源性信号，实现对BLPNs分解和前药活化的选择性可调控制，进一步证明了BLPNs在肿瘤中的特异性分布和高效肿瘤免疫治疗中免疫调节剂的刺激激活作用。本研究结果为肿瘤药物特异性输送和精准的癌症免疫治疗开辟了新的途径。
本研究使用欣博盛IL-6, TNF-ɑ和IFN-γ ELISA试剂盒检测了小鼠肿瘤组织样本中的细胞因子的含量。
